C. elegans: A gateway to aging research

Caenorhabditis elegans is a key tool in aging research, offering a short lifespan and genetic similarities to humans for rapid study of healthspan and lifespan interventions. This model is essential in unraveling the biological mechanisms of aging, identifying compounds and pathways that can delay age-related decline.

As a whole organism, C. elegans provides a more integrated system than cell cultures, while avoiding the ethical and logistical challenges of vertebrate models.

From research to results: Our approach

  1. Synchronized populations of C. elegans (L4 larval stage) are injected into the SydLab™ One platform (day 0)
  2. SydLab™ One tightly controls the liquid environment to which each worm is exposed, including the compounds to be tested.
  3. Images or videos are acquired via time-lapse microscopy at the desired frequency.
  4. Our AI-based algorithm precisely measures all aging-related parameters for each individual C. elegans, and provides a detailed analysis of the entire population.
  5. The experiment ends when no nematodes are alive.

Case Study: Lifespan and Healthspan analysis

We evaluated the effects of Paraquat and Nicotinamide (NAM) treatments against control conditions using two approaches: Nagi Bioscience’s fully automated SydLab™ One platform and traditional manual annotations.

The SydLab™ One platform accurately quantifies the expected lifespan changes in C. elegans subjected to varying treatments. It demonstrates the anticipated lifespan shortening with increasing concentrations of Paraquat and the lifespan extension with higher doses of Nicotinamide. The results obtained with the SydLab™ One platform are consistent with those obtained through manual plate-based assays, while increasing reproducibility and reducing time-to-results.

To delve deeper, we investigated the effects of varying levels of caloric restriction on
C. elegans.

The SydLab™ One platform quantified the expected lifespan extension resulting from progressive food dilution interventions (A), aligning with existing literature (66% of ad libitum: +10%; 33% of ad libitum: +39%). Phenotypic analysis reveals that both food dilution interventions significantly enhance motility (B), a hallmark of increased healthspan, while negatively impacting reproduction and worm size (C). Furthermore, food dilution significantly preserves intestinal integrity during aging, as evidenced by a higher intestine diameter-to-body diameter ratio (D).
